Protocol Development for In vitro Propagation of Enset [Ensete ventricosum (Welw.) Cheesman] Clones Using Shoot Culture

Ensete ventricosum is a species in the Musaceae family. Through conventional propagation, it is impossible to satisfy the sucker requirement. Hence, this research was initiated to develop a protocol for in vitro propagation of enset clones. For initiation, MS medium with different levels of BAP were used. Days of initiation and initiation percentage was recorded. For multiplication, combination of BAP and NAA in different levels were used. The number of shoots, leaves and shoot length were recorded. Different levels of IBA were used for rooting. Root length, numbers and rooting percentage were recorded. All experiments were done in CRD. BAP concentrations were significant for days to initiation and initiation percentage. Earlier shoot initiation was recorded on MS medium with 3.75 mg/l BAP. Maximum initiation percentage was obtained from a medium with 2.5 mg/l BAP. Eight and seven days were the minimum date of shoot initiation. The combination of BAP and NAA significantly influenced multiplication. Medium with 2.5 mg/l BAP +1.5 mg/l NAA gave five shoots/explant. Highest shoot length was 5.83 and 5.87 cm. MS medium + 1.5 mg/l IBA gave 5.33 roots and 5.17 cm root length as well as rooting percentage of 95% in both clones. Among the acclimatized, 92 % survived.
