Taro crop [Colocasia esculenta L. (Schott)] is belongs to Araceae family. The main objective of this study was to study the effect of three culture media viz; MS, N6 and B5 on in vitro growth of C. esculenta cv. ‘Boloso I.’ All the media supplemented with 8mg/L BAP and 3 mg/L IAA for multiplication of shoots. Comparison of in vitro rooting was performed by supplementing all the three media with 0.5 mg/L IAA. Highest shoot proliferation percentage (93.33%); maximum shoot number/explants (6.75); leaves number/explants (3.20); shoot length/explants (5.36cm) and leaf length/explants (2.13cm) were obtained on MS medium. In rooting stage, ½ MS medium supplemented with 0.5 mg/L IAA gave maximum rooting percentage (100%); highest number of root/explants (6.0) and longest root/explants (8.20cm). In all parameters, Chu (N6) medium showed poorest in taro propagation and B5 medium came in between N6 and MS medium. Analysis of data showed that there was a high significant difference between the three culture media. The strength of culture media also highly affects the in vitro rooting of taro. In conclusion, MS medium is the most suitable medium for in vitro production of C. esculenta cv. Bolosso I.
